3/30/2023 0 Comments Ubar ar15 lower![]() ![]() Next, sperm quality of frozen-thawed semen immediately after thawing was measured at 37 C by swim-up sperm count, sperm motility, spermatozoa with an intact acrosome and migration in polyacrylamide gel and then compared with the fertility of the semen used for artificial insemination. In ejaculates from six bulls, the number of spermatozoa swimming up was related to the number of live cells present (R2 = 0.97). In an initial test of the swim-up procedure, non-frozen sperm samples with different ratios of live to dead cells were prepared and tested for the number of spermatozoa counted by the swim-up procedure. The ability of the Percoll gradient method to extract motile spermatozoa was higher than that of the swim-up technique (20 +/- 15.6% versus 0.8 +/- 13.6%). Percoll gradient preparation produced a significantly higher final concentration of spermatozoa than swim-up preparation (mean +/- SEM: 6.6 +/- 1.5 x 10(6)/ml versus 1.9 +/- 0.2 x 10(6)/ml P less than 0.01) but a significantly lower sperm motility (69 +/- 2% versus 94 +/- 1% P less than 0.001) and a lower number of normal forms (55 +/- 2% versus 64 +/- 2% P less than 0.01). The oocytes for each trial were distributed at random between the two sperm preparations and incubated with the same number of motile spermatozoa. Each semen sample was prepared by both a swim-up technique and a simplified discontinuous (50%, 70%, 90%) Percoll gradient. Sixty trials of in-vitro fertilization (IVF), 38 with normal semen and 22 with abnormal semen, comprising 734 oocytes were included in the study. En este estudio se observó que la motilidad espermática no influyo en la tasa de fertilización y se concluye que ambos métodos de capacitación espermática son aptos para la producción de embriones in vitro. La producción total de embriones con ambos métodos fue de 95 embriones transferibles, con una tasa de desarrollo de 21,7%. Los resultados indicaron que la mayor concentración espermática y tasa de fertilización se observaron con Swim up (4,8 x 10 6 y 83,7 ± 7,8 %, respectivamente), mientras que con el Gradiente de Percoll se lograron valores de 3,5 x10 6 y 60,22 ± 6,3% para concentración espermática y tasa de fertilización. La concentración espermática se midió utilizando un SpermaCue® y la tasa de fertilización se evalúo a las 18 horas post fertilización utilizando microscopio con aumento de 100x. Para la separación de la fracción espermática motil se utilizaron los métodos Gradiente de Percoll® y Swim up. Transcurrido este tiempo se procedió a la FIV. Posteriormente, se seleccionaron ovocitos con más de cinco capas de células del cúmulo y se colocaron a madurar por 20 horas. ![]() Para ello se utilizaron ovocitos provenientes de ovarios post mortem que fueron transportados al laboratorio donde se puncionaron los folículos antrales (entre 2 mm y 5 mm). Con el objetivo de examinar el efecto que tiene el método de capacitación sobre la concentración y la tasa de fertilización se realizó este trabajo en el Laboratorio de Biotecnología del IDIAP. Un primer requisito en el proceso de preparación de los espermatozoides para la fertilización in vitro (FIV) bovina incluye la separación de los mismos del diluyente y/o del crioprotector. RESUMEN Los espermatozoides mamíferos no son capaces de fertilizar el ovocito inmediatamente primero deben sufrir un periodo de preparación llamado capacitación espermática, que se define como el conjunto de modificaciones que ocurren después de la maduración en el epidídimo, y que le confieren la capacidad de fertilizar al ovocito. ![]()
